Yes, you can enable weekends as operational days for both Prime and non-Prime offers. Enabling weekends as operational days is mandatory for Prime offers. You can set your weekend operation settings for all offers (by selecting Shipping All - Prime and Non-Prime Offers) or only Prime offers (by selecting Shipping Prime Offers Only) under General Fulfillment Settings and Fulfillment Settings for Shipping Region Automation.
By default, the setting for both Saturday and Sunday operations is set to Not Shipping. This means weekend operations is OFF. Changing from Not Shipping to any other option from the dropdown means you are turning ON weekend operation. By turning ON this setting, these days will be counted towards the Expected Ship Date and the Estimated Delivery Date. Please ensure you ship on time as this might impact your Late Shipment Rate.
Customers will see a cut-off time one hour prior to the one you set to allow for order processing. For example, if you have set the cut-off time for Next Day shipping as 3:00 p.m., customers see the cut-off time as 2:00 p.m.
If you are using Shipping Region Automation to define your delivery regions, order cut-off times can be adjusted by changing the last pick-up time for the shipping service. Once the last pick-up time is set for the shipping service, sellers can continue to adjust the cut-off time within a certain time window. You will be able to set the cut-off time for the whole week or by every day of the week.
If you are not using Shipping Region Automation, you will still be able to set order cut-off times for your products. However, you cannot set cut-off times by shipping service and day of the week.The cut-off time set under General Fulfillment Settings will be applied for all operational days (including weekends if enabled).
The last carrier pick-up time on the Order Fulfillment Settings acts as a mechanism for you to set more precise order cut-off times. You will not be able to set an order cut-off time for a shipping service that is later than the last pick-up time. The pick-up time is not used for the calculation of the promised delivery date shown to customers.
We understand that there might be multiple shipping services picking up packages on a given day. For example, Yamato and Japan Post might have a pick-up time of 5:00 p.m. and 6:00 p.m. respectively. Configuring Order Fulfillment Settings (under Fulfillment Settings for Shipping Region Automation) accordingly will allow you to receive orders up to a cut-off time of 4.30 p.m. and 5.30 p.m. respectively, assuming the order cut-off time is set 30 minutes before carrier pick-up time. Ensure that you have sufficient time between your order cut-off time and carrier pick-up time.

You can configure cut-off time and last pick-up time by shipping service only if the shipping service is being used to define your shipping regions on your automated shipping template.You cannot disable a shipping service on Order Fulfillment Settings except over the weekend. You cannot disable a shipping service on Order Fulfillment Settings except over the weekend. Note that, if weekend operations have been turned on, you must have at least one shipping service selected that picks up on the weekend.
If you have selected multiple shipping locations on your automated shipping templates, the promised delivery date will be automatically adjusted based on the customer location, time of day, and the time it takes for each shipping service to deliver to the customer location from each of the shipping locations. After evaluating all these factors, we will display the best delivery promise to the customer. This means that you may continue getting orders with an expected ship day for the same day until the time is past the order cut-off times of all your preferred shipping services.
If you have turned on Saturday or Sunday as an operating day, you are expected to ship on the selected day and ensure that the carrier picks up and scans on that day. For Prime offers, you are required to ship on both Saturday and Sunday. If the carrier is unable to pick up and scan the package on Saturday or Sunday, you have to select other/additional carriers and ship services to support it.
If an order is placed before your cut-off time you must ship the item on the same day. However, if an order is placed after cut-off time, you can ship the item on the next business day. If Saturday is enabled as an operating day, then orders received after cut-off time on Friday are expected to be shipped out by Saturday.
